Ames test는 bacterial test organism을 이용하여 mutagen을 가려내는, 간단하면서도 비용이 많이 들지 않는 방법이다. 그래서 Ames test에서는 bacterial system이 mammalian test system에 더 근접하도록 하기 위해 activating enzyme의 source로서 liver homogenate S-9를 넣어준다. Chemical의 mutagenic effect는 종종 organism의 enzymatic pathway에 의해 influence을 받아 nonmutagen이 mutagen으로 전환되거나 혹은 반대로 mutagen이 nonmutagen으로 전환되기도 하는데, 포유류에서는 이러한 toxification이나 detoxification이 간(liver)에서 일어난다. 이 균주는 back-mutation이 일어나 his+로 바뀌지 않는 한 histidine이 없는 배지에서 자라지 못할 것이다.
